Abstract

Recent advancements in marker development and the sequencing of the human genome have enabled the mapping and identification of genes for single locus disorders to proceed at a rapid pace. Attention is now focused increasingly on how to identify genes for common, complex diseases. Twin, adoption, and family studies support the hypothesis of strong (but not complete) genetic components for disorders such as schizophrenia, manic depression and noninsulin dependent diabetes mellitus. Nonetheless, segregation analysis and linkage analysis have not yet proven successful in delineating homogeneous subtypes or in identifying specific genes which contribute to susceptibility in the majority of cases.
