Abstract

The use of hydrogen/deuterium (HD) exchange coupled with mass spectrometry (MS) for the study of the binding of Mg (II) to ras-GDP protein was presented. The H/D exchanged solution was prepared by quenching the stock ras-GDP-Mg solution with decreased pH of 2.6. The complex dissociated to apo-ras, leaving an H/D exchange pattern that represented the complex. the dissociation constant was measured by titration of ras-GDP with Mg 2+.
