Single-molecule super-resolution imaging of molecular orientation using a tri-spot point spread function

Abstract

We demonstrate the use of an engineered Tri-spot point-spread function and a joint sparse deconvolution algorithm for imaging the positions and 3D orientations of single molecules. Our images reveal nanoscale order and disorder in DNA strands.
