Rapds and simple sequence repeats detect genetic variation in apple (malus x domestics borkh.) cultivars and roütstücks

Abstract

DNA amplification fingerprinting using arbitrary decamer primer(s) and simple sequence repeat primer(s) was applied to find genetic relationship and molecular characterization of apple cultivars and rootstocks. 2O apple cultivars and 10 apple rootstocks were screened for polymorphic RAPD studies uith 35 arbitrary 10-mer primers and 21 simple sequence repeat primers. Among apple cultivars and rootstocks a total of 68 and 55 bands with arbitrary primers and 6O and 73 bands with SSR primers were observed, r espec11veI y. Out of that 41 and 33 bands with arbitrary primers and 43 and 56 bands with SSR pr lmers were polymorphic. We have developed a DNA fingerprinting system based on the informative RAPD loci amp 1 if led by both the primera, that allowed the rapid identification of apple cultivars and rootstocks. Cluster analysis of SM and J coefticients based on UPGMA was carried out and dendrogram was constructed.
