Localizing and tracking single emitters in three dimensions using a double-helix point spread function

Abstract

Single fluorescent emitters can be localized and tracked in 3D with less than 1000 detected photons by using a DH-PSF. The tracking of quantum dots both in solution and inside a living cell is demonstrated.
