High-throughput blood analysis on a chip using lensless digital holography

Abstract

We present a lensfree holographic microscopy platform toward high-throughput on-chip blood analysis, which may facilitate point-of-care diagnostics in resource-poor environments. In this lensfree imaging modality, whole blood samples are illuminated with a spatially incoherent light source to record holographic diffraction patterns of cells on a digital sensor array. Through digital processing of the recorded lensfree holographic images, automated counting of red blood cells (RBCs) at densities up to ∼0.4 Million cells/μL is performed, along with characterization of the cell volume histogram and hemoglobin density of RBCs in whole blood. We further demonstrate automated counting of white blood cells using the same platform, also yielding sub-cellular spatial resolution to differentiate granulocytes, monocytes and lymphocytes from each other.
